Antarktika kaynaklı örneklerden dalapon herbisitini parçalayan bakterilerin izolasyonu, karakterizasyonu ve tanısı

Dalapon with active ingredient 2,2-dichloropropionic acid is a widely used herbicide variety compared to its alternatives. Although it is used effectively against weeds, these and other herbicides both cause stress in the plant and harm the human and other beneficial forms of living by accumulating in the soil and mixing with the groundwater. Studies for detoxification of bacteria in heavy metals and disintegration of pesticides are carried out by many researchers. In this study, it was aimed to isolate bacteria that break down the dalapon herbicide from Antarctic samples, to identify these bacteria with molecular meaning, to characterize them in morphological and biochemical tests, and to investigate the usability of isolates as a bioremediation agent in agricultural areas. The samples used for this purpose were delivered to us as soil and sea water samples collected from the Western Antarctic Peninsula during the 3rd Turkish Antarctic Science Expedition. Psychrophilic bacteria isolate K and L, which can grow in minimal nutrient media containing 20 mM 2,2-dichloropropanoic acid as a single carbon source are isolated for the first time from the seawater sample belonging to Galindez Island of Antarctica. The nucleotide sequences obtained as a result of 16S rRNA analysis were compared to the 16S rRNA sequences of other bacteria with the BLAST tool in the NCBI database and it was found that the isolates were highly similar to the species belonging to the genus Psyhcrobacter. The 16S rRNA gene sequences of the isolates K and L were deposited to GenBank as Psyhcrobacter sp. strain TaeBurcu001 and Psychrobacter sp. strain TaeBurcu002, respectively. The isolates differ from other Psyhcrobacter species in the literatüre in terms of some morphological and biochemical characters. As a result of the pathogenicity test, Psychrobacter sp. strain TaeBurcu001 and Psychrobacter sp. strain TaeBurcu002 have been shown to be used as bioremediation agents in agricultural areas contaminated with dalapon.
